Abstract

In veterinary practice developed various methods of cryopreservation of animals` red blood cells. In our previous studies have shown the effectiveness of dimethylsulfoxide (DMSO) for cryopreservation of horses erythrocyte, but after low temperature storage cells should not only intact, but functionally complete. Various factors of low action can change the speed of biochemical reactions in cells, which may influence on their function and influence of DMSO on membrane transport systems can be crucial for cell survival under stressful conditions. To study changes in the activity of Ca2+-ATPase horse red blood cells under the influence of different concentrations of DMSO cryopreservation and factors. The activity of Са2+-АТPase in saponin-perforated red blood cells of horse in the process of cryopreservation with the different concentrations of DMSO has been studied. It has been shown that the high concentrations of DMSO (15%) inhibited the activity of ATPase, but low concentrations of cryoprotectants (5-10 %) did not influence the work of the enzyme. The activity of Са2+-АТPase of horses erythrocytes in the process of cryopreservation and cryoprotectant elimination do not trustworthy change as compared with control one. Modification of the activity of Ca2+-ATPase in the influence of DMSO and low temperatures indicates the participation of Ca2+ and systems that direct their level. Conclusions and prospects for further development. Cryopreservation of horses erythrocytes with DMSO accompanied by changes in the activity of Ca2 + - ATPase. Changes in the activity of Ca2+ - ATPase expressed using relatively high concentrations of dimethylsulfoxide. Violation of enzyme activity is reversible and disappears when you remove cryoprotectant from cells. Thus, further research activities Ca2 + -ATPase in cells that come under the influence of DMSO and low temperatures will help develop ways to stabilize and adaptation of cells to stress action.
